Summary A thorough corneal and anterior segment examination relies on slit-lamp biomicroscopy; however, that type of examination cannot fully answer some clinical questions. Biopsy and tissue sectioning may be needed to provide additional information about corneal structures and corneal pathology and to examine portions of the anterior segment not readily visible with the slit lamp. While in vitro sectioning, as in histologic preparations, serves that purpose, the practical need exists for in vivo "optical sectioning" in the clinic. A variety of anterior segment imaging modalities serve this purpose. Choosing the right test or device can be a crucial aid in making the correct diagnosis
